To precisely quantify sugar levels in solutions in real time, a phase lock-in rotating analyzer (PLRA) polarimeter has been developed and implemented. The incident beams, exhibiting polarization rotation, caused a phase shift in the sinusoidal photovoltages of the reference and sample beams, which were detected by the two spatially separated photodetectors. The monosaccharides fructose and glucose, and the disaccharide sucrose, have been quantitatively determined, revealing sensitivities of 12206 deg ml g-1, 27284 deg ml g-1, and 16341 deg ml g-1 respectively. From the fitting functions, respective calibration equations were generated for determining the concentration of each individual dissolved substance in deionized (DI) water. Considering the predicted results, the absolute average errors in the readings for sucrose, glucose, and fructose stand at 147%, 163%, and 171%, respectively. In freeze-dried pharmaceutical products, residual moisture (RM) is a vital critical quality attribute (CQA) that needs close monitoring because it substantially impacts the stability of the active pharmaceutical ingredient (API). The Karl-Fischer (KF) titration, a standard experimental method for RM measurements, is destructive and time-consuming in nature. Consequently, the use of near-infrared (NIR) spectroscopy has been studied extensively in the last decades as an alternative method to measure the RM. This paper introduces a novel NIR spectroscopy-based machine learning approach for predicting RM levels in freeze-dried products. Employing a linear regression model alongside a neural network-based model, two distinct modelling strategies were examined. Careful selection of the neural network's architecture was undertaken to ensure accurate residual moisture prediction by minimizing the root mean square error against the learning dataset. Moreover, visual evaluations of the results were achieved through the presentation of parity plots and absolute error plots. During the development of the model, the encompassing wavelength spectrum, the spectral shapes, and the model's type were meticulously evaluated. To explore the prospect of a model derived from a single product, applicable to a broader array of products, was a key part of the investigation, and the performance of a model trained on multiple products was also studied. Analyses of diverse formulations revealed that the majority of the dataset contained varying percentages of sucrose in solution (3%, 6%, and 9% specifically); a smaller proportion involved mixtures of sucrose and arginine at different concentrations; and a single formulation included trehalose as an alternative excipient. The model, created for the 6% sucrose mixture, proved reliable in predicting RM in various sucrose solutions, even those including trehalose, but its reliability diminished in datasets containing a higher proportion of arginine. In conclusion, a model encompassing the entire world was built by incorporating a specific percentage of the total dataset into the calibration phase. In this paper, the results presented and discussed show that the machine learning model's accuracy and robustness surpass those of linear models.

Our research aimed to pinpoint the molecular and elemental alterations in the brain characteristic of early-stage obesity. A combined methodology utilizing Fourier transform infrared micro-spectroscopy (FTIR-MS) and synchrotron radiation induced X-ray fluorescence (SRXRF) was adopted to determine some brain macromolecular and elemental parameters in high-calorie diet (HCD)-induced obese rats (OB, n = 6) and their lean counterparts (L, n = 6). Studies demonstrated that the administration of HCD resulted in changes to the lipid and protein makeup and elemental composition of essential brain regions responsible for energy homeostasis. Obesity-related brain biomolecular aberrations, as evidenced in the OB group, were characterized by increased lipid unsaturation in the frontal cortex and ventral tegmental area, elevated fatty acyl chain length in the lateral hypothalamus and substantia nigra, and a reduction in both protein helix-to-sheet ratio and the percentage fraction of turns and sheets in the nucleus accumbens. The study also revealed that particular brain components, such as phosphorus, potassium, and calcium, showcased the most significant difference between the lean and obese groups. HCD-driven obesity results in tangible structural alterations within lipids and proteins, as well as redistributions of elemental components in brain areas essential for energy maintenance. The application of X-ray and infrared spectroscopy in a combined fashion was proven a dependable means of identifying elemental and biomolecular changes in rat brain tissue, thereby improving our knowledge of the intricate connections between chemical and structural processes involved in appetite regulation.

Environmentally benign spectrofluorimetric techniques have been applied for the determination of Mirabegron (MG) in both pure drug and pharmaceutical formulations. Employing Mirabegron as a quencher, the developed methods depend on fluorescence quenching of tyrosine and L-tryptophan amino acid fluorophores. The experimental procedures for the reaction were examined and enhanced for optimal results. MG concentration, ranging from 2 to 20 g/mL for the tyrosine-MG system at pH 2 and from 1 to 30 g/mL for the L-tryptophan-MG system at pH 6, demonstrated a direct proportionality with the corresponding fluorescence quenching (F) values. The ICH guidelines served as the basis for the method validation. For the determination of MG in the tablet's formulation, the cited methods were used in a sequential manner. Regarding t and F tests, the results from the cited and referenced methods display no statistically significant difference.
